IP -mediated IP  release drives endothelial Ca   waves

En face mesenteric artery preparation stained with Cal-520/AM (5 µM, grey) and cIP3 (5 μM). Ca activity was stimulated by localized photolysis (yellow circle) after baseline recording (10 Hz; baseline cut short for the purpose of video). Localized U73122 application (10 µM; 15 mins) was performed using a puffer pipette (location denoted by yellow arrow) in the presence of counter flow. The resulting Ca   activity is visualized in green (Control) and purple (15 mins focal U73122 application). Signal intensity profiles across the centre horizontally and diagonally are shown, along with the surface plot for the recording. Scale bar: 50 μm.
