IP -generated Ca   waves are able to cross endothelial discontinuities

En face mesenteric artery preparation stained with Cal-520/AM (5 µM, grey) and cIP (5 μM). Ca activity was stimulated by localized photolysis (yellow circle) after baseline recording (10 Hz; baseline cut short for the purpose of video). Control cIP -induced Ca   activity was first generated, then an endothelial discontinuity created using a puffer pipette with a fire-polished tip. Preparations were left to rest for at least 15 mins after the endothelial discontinuity had been created, and a subsequent cIP -induced Ca   activity generated. Scale bar: 50 μm.
